The war on cancer is now entering its 4th decade. Current treatments such as surgery, chemotherapy and radiation therapy are not always adequate, and there is a need for newer technologies. Nanotechnology has shown great promise in cancer therapy, with more targeted and specific drug delivery.

Nanoscale devices can easily enter most cells, and those < 20 nm can move out of blood vessels as they circulate through the body. They can serve as customized, targeted drug delivery vehicles to carry large doses drugs into cancerous cells, without affecting healthy cells.

Some drugs that have previously failed clinical trials can also be reexamined using nanotechnological approaches. Drugs can be made more soluble, kept in circulation for longer periods, and their stability can also be increased tremendously by encasing them within nanosized drug carriers.

In case of brain cancers, many drugs have difficulty in crossing the blood– brain barrier to attack the tumor. Drug-loaded nanoparticles are able to penetrate this barrier, and have been shown to greatly increase therapeutic concentrations of anticancer drugs in brain tumors.

This image is a 3D reconstruction of a hollow multi-cellular prostate tumor spheroid (cluster of cells) imaged by confocal microscopy. Cell nuclei are stained red, and the nano drug carriers entering cells are shown in blue. These carry the cytotoxic drug doxorubicin (black and red beads inside the dendrimer). Doxorubicin encapsulated inside the dendrimer is able to penetrate deeper into the spheroid better killing the prostate tumor cells.

3D cultures, a model for cancer research.

Pictured here is a spheroid of hepatocellular carcinoma (liver cancer) cells. This is a model scientists use for studying cancer growth. Conventionally such cells are grown and studied in a 2D monolayer (on a dish) where it is easy to see the different structures within each cell. However, cancerous 3D spheroids such as these provide researchers with accurate depictions of tumor environment in the body, in a way a more realistic model considering our bodies are 3D structures. This model is helpful to study drug delivery to different parts of the tumor and how is that the tumor responses to such treatments. These invaluable tools are becoming more commonplace in labs across the world and represent an important technology for cancer research.

With 3D cell culture becoming more common, there is a need to develop ways of imaging these spheroids accurately. Visikol® has developed a novel approach to image 3D spheroids using a confocal microscope.

Born out of labs from Rutgers University, team members of Vikisol developed a clearing agent to visualize plant tissues which are normally thick and opaque. This technology was then adapted to 3D cultures and commercialized for animal pathology.
