So first session back in the gym today using my puregym 7 day free pass (thank you @_amyfitness_ 💖💖). It started off well with 10 mins cardio then started legs with a superset which was fine but I was more out of breath than normal. Came to squats and legit almost passed out so had to cut my workout short ☹️ did 20 mins of LISS at the end instead tho which was good! Hoping tomorrow will be better 🙂

Potency analysis is probably the most widely known term in the cannabis testing industry, but what is it? As I have mentioned before, potency analysis is used to quantify the amount of cannabinoids present in a sample, sometimes this means measuring as little as 5mg (aka five one-one thousandths of a single gram) of cannabinoids in a 50 gram sample, and other times it means handling distillate concentrates that are often over 99% cannabinoids.
With such a wide variety of sample types and concentrations,several methods for sample preparation had to be developed. In today's picture I have included several types of marijuana products listed left to right, top to bottom, marijuana flower, distillate, CO2 extract, shatter, rso, after dinner mints, Taffy, and emulsified extract.
Analytical 360 looks for and quantifies 7 of the most common cannabinoids, cbda, thca, cbg, cbd, cbn, thc, and cbc.
So What kind of instrumentation do we use to measure the content of cannabinoids? Find out tomorrow!

Yesterday's question-How are e. coli and Salmonella confirmed to be present?
Answer-it really depends on the method of microbial analysis. Many labs in Washington state are utilizing polymerase chain reaction (PCR, many of my biochemistry brothers and sisters are scoffing because they know how unreliable this technique can be for quantitative analysis) for all of their microbiological analysis. Luckily, here at analytical 360 we utilize plates. For e coli and salmonella we use selective broth and selective plates to display the presence of e. coli 0157 (the bad kind, and yes there is way more than just one kind of e.coli) and salmonella. If these plates confirm that these bacteria are present, a final antibody test is used to confirm that the specific bacteria is present next to a positive control for comparison. In this antibody test, the antibody is attached to a silica bead, when several of these attach to cells the silica beads cluster together and give a positive reading for the presence of the specific protein found on the surface of the bacteria (that is the only thing the antibody can attach to). The picture attached to the today's message is of some of these plates, the white plate select for selects for E coli. The red plate selects for Salmonella. I have also included a second image which is the antibody test for e. Coli 0157.
Believe it or not there is still a lot that we can say about Micro so we will come back to the subject, however for now I just wanted to touch on this testing and give this some time to sink in. Later we will look at some results and make conclusions on some of the factors that may have led to specific results.

Yesterday's question-what are the various types of products that need microbial testing and what are the limits for the various bacteria and yeast and mold?
Answer-Washington State has three categories of products that require microbial testing unprocessed material (buds, flower,etc. shorthand- upm), processed material (keif, shorthand- pm) and solvent-based extracts (bho,rso,pho, shorthand-sbe). I will list the allowed limits for each below
Total aerobic plate count upm-100,000
Pm-100,000
Sbe-10000
Yeast and mold
Upm-10000
Pm-10000
Sbe-1000
Bile tolerant gram-negative bacteria
Upm-1000
Pm-1000
Sbe-100
Total coliforms

e-coli and salmonella are not allowed to be present in any samples, if these are detected in any amount, the whole lot must be destroyed. In addition to all of this there are retest limits, the retest limit is a multiple of the maximum allowance of cfu per gram (the numbers listed above) with total aerobic count being two times the upper limit yeast and mold being five times the upper limit coliforms being two times the upper limit and bile tolerant gram-negative being two times the upper limit.

Microbial testing requires meticulous sterile technique which is the hardest part imo. The cfu/g is calculated by counting the colonies on the plates and multiplying by the dilution factor (ya know, math). In today's picture we have a bunch of counted plates ready for disposal. So if we determine all the cfu/g by counting the plates, how do we determine if salmonella and ecoli are present?... find out tomorrow!
